Isolation of exosomes and extracellular vesicles used to be a process involving ultracentrifugation which is a cumbersome and non-standardized process. At Exosome Diagnostics, we have emphasized practical routine use in clinical laboratories during our product and protocol development. The ExoLution range of exosome isolation products provides simple bind-wash-elute protocols that are easy to execute and reproducibly yield high quality material.
Targeted sequencing and whole transcriptome sequencing
Currently, most clinically actionable RNA biomarkers are messenger RNAs (mRNAs) and long non-coding RNAs (lncRNAs). However, profiling of exosomal RNA has been mostly limited to small RNAs which is a major limitation for biomarker discovery. At ExosomeDx, we have developed a highly reproducible and extremely sensitive long RNA-Seq assay (patent pending) optimized to profile long RNAs in exosomes. Our long RNA-Seq assay routinely detects over 10,000 mRNAs and thousands of lncRNAs that has previously remaining undetectable in exosomes; therefore, tremendously expanding the scope of biomarker discovery.
Biomarker discovery/dedicated bioinformatics pipelines
If the list of potential biomarkers has already been identified, targeted sequencing of a panel of genes of interest offers additional advantages of deeper sequencing, reduced sequencing noise and increased sample multiplexing over whole transcriptome sequencing. At ExosomeDx, we have developed robust and highly sensitive assays for targeted expression profiling of gene panels of various sizes from exosomal RNA derived from subjects' biofluids.
Dedicated NGS that are built to analyze exosomal RNA
Exosome Diagnostics has developed highly scalable and secure analysis pipelines that are optimized for analyzing nucleic acid content from exosomes. This includes modules for analyzing RNASeq data as well as highly sensitive mutation detection for targeted sequencing applications. We also offer machine learning pipelines to identify biomarkers for disease identification/stratification from disparate kinds of data.
